Mechanism of Action

Darunavir is a second-generation, non-peptidic HIV-1 protease inhibitor that prevents the maturation of infectious virions by blocking the proteolytic cleavage of viral polyproteins. HIV-1 protease is a homodimeric aspartyl protease essential for processing the Gag and Gag-Pol polyprotein precursors into functional structural proteins and enzymes — namely the matrix, capsid, nucleocapsid, reverse transcriptase, integrase, and protease itself. Darunavir binds with exceptionally high affinity to the active site of HIV-1 protease, forming extensive hydrogen bond interactions with the backbone atoms of the enzyme, particularly at the Asp25-Asp25' catalytic dyad and the flap region. The dissociation constant of the darunavir-protease complex is approximately 4.5 x 10⁻¹² M, which is 100 to 1000 times lower than that of first-generation protease inhibitors, reflecting the extraordinary binding strength of this molecule. Darunavir was specifically designed to fit tightly within the substrate envelope of HIV-1 protease, meaning it occupies the same three-dimensional space as the natural peptide substrate.
