GlibenclamideThird Party Manufacturing in India
Looking to get Glibenclamide manufactured under your brand? Our WHO-GMP certified partner facilities support PCD franchise partners and global export opportunities structured across 25+ countries.
Mechanism of Action
Glibenclamide works by stimulating insulin secretion from pancreatic beta cells. It binds to the sulfonylurea receptor type 1 (SUR1) subunit of ATP-sensitive potassium (K-ATP) channels on the beta cell membrane. This binding causes the channel to close, preventing potassium ions from leaving the cell. The resulting membrane depolarization opens voltage-dependent calcium channels, allowing calcium ions to enter the cell. The rise in intracellular calcium concentration triggers the exocytosis of insulin-containing secretory granules, releasing insulin into the bloodstream.
As a second-generation sulfonylurea, glibenclamide has a much higher affinity for the SUR1 receptor compared to first-generation agents like chlorpropamide and tolbutamide. This allows for lower therapeutic doses and improved tolerability. The insulinotropic effect of glibenclamide is glucose-dependent — it is more effective when blood glucose levels are elevated and less likely to cause hypoglycemia when glucose levels are normal. Glibenclamide may also have mild extra-pancreatic effects including improved peripheral insulin sensitivity and reduced hepatic glucose production.
Therapeutic Uses & Indications
Therapeutic Uses
Glibenclamide is prescribed for the management of type 2 diabetes mellitus:
- Type 2 Diabetes (Monotherapy): For patients who cannot achieve glycemic control through diet and exercise alone
- Combination Therapy with Metformin: Widely used fixed-dose combination for enhanced glycemic control
- Combination with Thiazolidinediones or Insulin: Used in advanced disease stages when beta cell function is partially preserved
- Gestational Diabetes: Used in some protocols when insulin is not feasible, though insulin remains the preferred agent
Why Manufacturers Choose Glibenclamide
Glibenclamide third party manufacturing in India is a well-established segment of the antidiabetic pharmaceutical market. Indian manufacturers produce glibenclamide tablets (2.5mg and 5mg) for PCD franchise networks, institutional supply, and export markets including Africa, Southeast Asia, and the Middle East. The molecule has been a cornerstone of diabetes management for decades and continues to see consistent demand.
For contract manufacturers, glibenclamide offers straightforward tablet formulation with excellent stability profiles. The API is readily available from multiple Indian sources, and WHO-GMP certified production lines can easily incorporate this molecule. With India's growing diabetes burden, the demand for cost-effective sulfonylurea therapy remains strong. Glibenclamide is also available in fixed-dose combinations with metformin, expanding its manufacturing and marketing potential for PCD franchise partners.
