Mechanism of Action

Aciclovir works by selectively inhibiting the replication of viral DNA.

It is a synthetic purine nucleoside analogue that requires activation by the viral enzyme thymidine kinase, converting it into aciclovir monophosphate. Host cellular enzymes then convert it into its active triphosphate form. This active form competitively inhibits viral DNA polymerase and incorporates itself into the growing viral DNA chain, leading to premature chain termination and stopping viral replication.
