Mechanism of Action

Doxazosin is a selective alpha-1 adrenergic receptor antagonist belonging to the quinazoline class. It works by competitively blocking postsynaptic alpha-1 adrenergic receptors located on vascular smooth muscle cells. This blockade inhibits the binding of norepinephrine, a naturally occurring catecholamine that normally triggers vasoconstriction. By preventing this interaction, doxazosin causes relaxation of both arterial and venous smooth muscle, leading to vasodilation and a significant reduction in systemic vascular resistance.
