Mechanism of Action

Fluorouracil works by blocking DNA and RNA synthesis to halt the growth of rapidly dividing cancer cells. It is a pyrimidine analog that acts as an antimetabolite. Once inside the cells, Fluorouracil is converted into active metabolites, including fluorodeoxyuridine monophosphate (FdUMP) and fluorouridine triphosphate (FUTP). FdUMP binds to and inhibits the enzyme thymidylate synthase, which blocks the methylation of deoxyuridylic acid to thymidylic acid. This action leads to a depletion of thymidine triphosphate, a necessary component for DNA replication and repair. Simultaneously, FUTP is incorporated into RNA, disrupting normal RNA processing and translation.

This dual disruption of nucleic acid metabolism selectively targets rapidly proliferating cells, triggering cell cycle arrest and apoptosis. Unlike non-specific cytotoxic drugs, Fluorouracil specifically targets cells in the S-phase of the cell cycle.
