Mechanism of Action

Gatifloxacin works by inhibiting two essential bacterial enzymes — DNA gyrase and topoisomerase IV. DNA gyrase is responsible for introducing negative supercoils into bacterial DNA, allowing proper replication and transcription. Topoisomerase IV is involved in separating replicated chromosomal DNA during bacterial cell division. By binding to these enzyme-DNA complexes, gatifloxacin prevents the re-ligation of DNA strands, leading to double-strand breaks and ultimately bacterial cell death.

As a fourth-generation fluoroquinolone, gatifloxacin exhibits enhanced activity against both gram-positive and gram-negative bacteria compared to earlier generation quinolones. The 8-methoxy group on its quinolone ring contributes to its improved activity against resistant strains and reduces the likelihood of resistance development. Gatifloxacin is particularly effective against respiratory pathogens including Streptococcus pneumoniae, Haemophilus influenzae, and atypical organisms like Mycoplasma pneumoniae and Chlamydia pneumoniae.
