LisinoprilThird Party Manufacturing in India

Mechanism of Action
Lisinopril works by competitively inhibiting angiotensin converting enzyme, a peptidyl dipeptidase that catalyzes the conversion of angiotensin I to the potent vasoconstrictor angiotensin II. By blocking this conversion, lisinopril reduces angiotensin II levels, leading to decreased vasopressor activity and lower blood pressure. The reduction in angiotensin II also decreases aldosterone secretion from the adrenal cortex, which reduces sodium and water retention and causes a modest increase in serum potassium. Unlike many other ACE inhibitors, lisinopril is not a prodrug and does not require hepatic activation, making it directly active after oral administration with predictable pharmacokinetics. It is excreted unchanged in the urine, which provides consistent drug exposure. Additionally, ACE inhibition prevents the degradation of bradykinin, a vasodilatory peptide that contributes to the overall blood pressure-lowering effect. The sustained suppression of the renin-angiotensin-aldosterone system also provides cardioprotective and renoprotective benefits beyond blood pressure control alone.
Therapeutic Uses & Indications
Therapeutic Uses
Lisinopril is prescribed for the treatment of:
- Hypertension: First-line therapy for mild to moderate hypertension, effective as monotherapy or in combination with thiazide diuretics and calcium channel blockers.
- Heart Failure: Adjunctive treatment in patients with congestive heart failure to reduce mortality and improve symptom control.
- Myocardial Infarction: Used post-heart attack to improve survival and reduce the risk of subsequent cardiovascular events.
- Diabetic Nephropathy: Provides renoprotective effects in patients with type 2 diabetes and early kidney disease.
Why Manufacturers Choose Lisinopril
Lisinopril is one of the most prescribed ACE inhibitors globally, with enormous volume across both branded and generic markets. Its once-daily dosing and excellent tolerability profile drive strong patient compliance and repeat prescriptions. Indian manufacturers benefit from established API supply chains and the availability of multiple tablet strengths from 2.5mg to 20mg, enabling broad product line coverage for PCD franchise partners and export to regulated markets. The molecule is cost-effective to manufacture and has a well-characterized stability profile, making it a staple product in any cardiovascular portfolio.
