Mechanism of Action

Losartan Potassium works by selectively blocking the angiotensin II type 1 (AT1) receptor, a key component of the renin-angiotensin-aldosterone system that regulates blood pressure and fluid balance. Angiotensin II, a potent vasoconstrictor, is formed from angiotensin I through the action of angiotensin-converting enzyme. When losartan binds to the AT1 receptor, it competitively and reversibly inhibits the binding of angiotensin II at this site. This blockade prevents angiotensin II from exerting its vasoconstrictor effects on vascular smooth muscle, its aldosterone-stimulating effects on the adrenal cortex, and its sodium-retaining effects on the kidneys. The result is vasodilation of both arterioles and veins, reduced peripheral vascular resistance, decreased aldosterone secretion, and enhanced sodium and water excretion, all contributing to sustained blood pressure reduction. Losartan is a prodrug that undergoes first-pass metabolism in the liver to form its active carboxylic acid metabolite, which is 10 to 40 times more potent than the parent compound. Unlike ACE inhibitors, losartan does not affect bradykinin breakdown, which explains the lower incidence of cough and angioedema associated with ARB therapy. Losartan also demonstrates renoprotective effects by reducing intraglomerular pressure and proteinuria, making it particularly valuable in diabetic nephropathy.

Therapeutic Uses & Indications

Therapeutic Uses

Losartan Potassium is an established angiotensin receptor blocker prescribed for cardiovascular and renal conditions:

  • Essential Hypertension: Losartan is indicated for the treatment of hypertension in adults and pediatric patients aged 6 years and older, either as monotherapy or in combination with other antihypertensive agents such as thiazide diuretics and calcium channel blockers.
  • Diabetic Nephropathy: Losartan delays the progression of renal disease in patients with type 2 diabetes and proteinuria, reducing the risk of progression to end-stage renal disease and the need for dialysis or kidney transplantation.
  • Stroke Prevention in Hypertensive Patients with Left Ventricular Hypertrophy: Losartan reduces the risk of stroke in hypertensive patients with left ventricular hypertrophy, an effect that is independent of its blood pressure lowering properties.
  • Pediatric Hypertension: It is approved for the treatment of hypertension in children aged 6 years and older, providing a well-tolerated first-line option for pediatric antihypertensive therapy.

Why Manufacturers Choose Losartan Potassium

Losartan Potassium is among the most commonly prescribed antihypertensive molecules worldwide and a top-selling ARB in India. Its availability at 25mg, 50mg, and 100mg tablet strengths creates a complete range of SKUs for PCD franchise and export markets. The molecule is included in the WHO Essential Medicines List and is covered under India's Jan Aushadhi scheme, ensuring consistent procurement volumes. Indian WHO-GMP certified manufacturers produce losartan tablets at competitive prices, serving both the domestic market and regulated markets across the US, UK, EU, and Australia.

This makes Losartan Potassium one of the most sought-after antihypertensive molecules for third party manufacturing in India, with consistent demand from PCD franchise operators and export-oriented companies.
