Mechanism of Action
Metronidazole works by entering bacterial and protozoal cells and interacting with DNA. Once inside the cell, the nitro group of metronidazole is reduced by bacterial nitroreductases to form reactive intermediates including nitroso radicals and hydroxylamine derivatives. These reduced metabolites disrupt the helical structure of DNA and inhibit nucleic acid synthesis, leading to cell death. This mechanism is selectively toxic to anaerobic microorganisms because only anaerobes and certain microaerophilic organisms possess the necessary reducing systems to activate the drug.
