Mechanism of Action

Mupirocin works by inhibiting bacterial protein synthesis. It specifically and reversibly binds to bacterial isoleucyl-transfer RNA (tRNA) synthetase, which is the key enzyme responsible for catalyzing the formation of isoleucyl-tRNA from isoleucine and tRNA. By blocking this critical enzyme, Mupirocin prevents the incorporation of isoleucine into growing bacterial proteins, which immediately halts protein synthesis and halts bacterial cell growth.
