Mechanism of Action

Ondansetron works by highly selectively antagonizing serotonin 5-HT3 receptors both peripherally on vagal nerve terminals and centrally in the chemoreceptor trigger zone (CTZ) of the area postrema.

By blocking these receptors, it prevents serotonin, which is released from the enterochromaffin cells of the small intestine during chemotherapy or radiation, from initiating the vomiting reflex. This targeted action effectively suppresses nausea and vomiting.
