Pyrazinamide works as a prodrug that requires activation by the pyrazinamidase enzyme produced by Mycobacterium tuberculosis. Once converted to its active form, pyrazinoic acid, it disrupts the bacterium's energy metabolism by interfering with fatty acid synthase I and membrane transport functions.
