Mechanism of Action

Serratiopeptidase is a zinc-containing metalloprotease enzyme belonging to the serralysin family (EC 3.4.24.40), isolated from the non-pathogenic enterobacterium Serratia marcescens. It exerts its therapeutic effects through the selective proteolytic degradation of non-living proteins and inflammatory mediators while leaving viable cells unharmed. The enzyme's active site contains a catalytic zinc ion coordinated by three histidine residues and a tyrosine residue, which facilitates the hydrolysis of specific peptide bonds. Serratiopeptidase reduces inflammation and edema by degrading key inflammatory mediators such as bradykinin, a peptide that promotes pain and swelling by increasing vascular permeability. By cleaving bradykinin and other kinins at the site of inflammation, the enzyme reduces local edema and pain. It also exhibits fibrinolytic activity by dissolving fibrin clots and denatured proteins that accumulate at sites of tissue injury. This helps in the clearance of dead tissue and promotes faster healing. Additionally, serratiopeptidase decreases neutrophil infiltration at sites of inflammation, thereby reducing the release of neutrophil-derived proteases like elastase that exacerbate tissue damage. Unlike non-steroidal anti-inflammatory drugs, serratiopeptidase does not inhibit cyclooxygenase enzymes, which means it provides anti-inflammatory effects without the gastrointestinal side effects associated with NSAIDs.
