Mechanism of Action

Ticarcillin + Clavulanic Acid is a combination antibiotic that works through two complementary mechanisms. Ticarcillin is a carboxypenicillin beta-lactam antibiotic that kills bacteria by preventing the cross-linking of peptidoglycan during bacterial cell wall synthesis. It binds to penicillin-binding proteins (PBPs) inside the bacterial cell wall, inhibiting the final transpeptidation step required for cell wall integrity, which ultimately causes bacterial cell lysis and death. Clavulanic acid is a beta-lactamase inhibitor that protects ticarcillin from enzymatic degradation. Many resistant bacteria produce beta-lactamase enzymes that cleave the beta-lactam ring of penicillins, rendering them inactive. Clavulanic acid acts as a suicide substrate, irreversibly binding to these beta-lactamases and neutralizing them. This allows ticarcillin to reach its target intact and exert its bactericidal effect against otherwise resistant organisms. The combination restores ticarcillin's activity against beta-lactamase-producing strains of bacteria including Pseudomonas aeruginosa, making it an important option for hospital-acquired and severe infections.
