Mechanism of Action
Tirofiban Hydrochloride is a non-peptide glycoprotein IIb/IIIa receptor antagonist that prevents platelet aggregation by blocking the final common pathway of platelet activation. Platelet aggregation is mediated by the binding of fibrinogen to the glycoprotein IIb/IIIa receptor on activated platelets. When platelets are activated by agonists such as ADP, collagen, or thrombin, the GP IIb/IIIa receptor undergoes a conformational change that allows it to bind fibrinogen. Each fibrinogen molecule can bind to GP IIb/IIIa receptors on two different platelets, forming cross-links that build a stable platelet thrombus. Tirofiban selectively and reversibly blocks the GP IIb/IIIa receptor, preventing fibrinogen binding and therefore inhibiting platelet aggregation regardless of the activating stimulus. This makes it one of the most effective antiplatelet agents available, with inhibition of up to 97% of ADP-induced platelet aggregation at therapeutic doses. Tirofiban has a rapid onset of action within 5 minutes and a short half-life of approximately 2 hours, with platelet function recovering within 4 to 8 hours after discontinuation.
